Thrombin is the active enzyme that splits fibrinogen into fibrin in final stage of blood coagulation. It is generated from its circulating inactive precursor, prothrombin, by the action of factor Xa aided by factor Va, phospholipid and calcium ions. Thrombin is a glycoprotein formed by a 6-kDa A chain linked by a disulfide bridge to a 31-kDa B chain which carries the serine protease catalytic site.

HYB 109-04 binds to human thrombin and to complex of human thrombin with the thrombin inhibitor hirudin. No cross-reaction is seen with prothrombin. Cross-reactivity with thrombin from other species has not been tested.

HYB 109-04 reacts strongly in ELISA with thrombin coated directly onto the microtiter wells. Biotinylation of thrombin blocks antibody binding.
